Born in 1828 at Charles Street in Mayfair, Charlotte Isabella Cust was the daughter of Lieutenant-Colonel the Honourable Peregrine Francis Cust and his wife Lady Isabella Mary née Montagu-Scott. She married John Clark-Kennedy on 8 September 1859. She died on 24 November 1914 at 6 West Eaton Place in London, leaving an estate valued at £38,790. 

According to a report in the Dumfries and Galloway Standard (2 December 1914): 'The funeral took place on Monday at Dumfries of the late Mrs John Clark-Kennedy of Knockgray, Kirkcudbright, who died in London last week. She was Charlotte Isabella, daughter of the late Colonel the Hon. Peregrine Cust by his marriage with a daughter of the fourth Duke of Buccleuch. In 1859 she became the second wife of the late Colonel John Clark-Kennedy of Knockgray, C.B., by whom she had three daughters. Her husband was the son of Lieutenant-General Sir Alexander Clark-Kennedy of Knockgray, who commanded the centre squadron of the Royal Dragoons at Waterloo, and with his own hand captured the eagle and colours of the 105th Regiment of French Infantry. In commemoration of this incident he received an honourable augmentation to his arms surmounted by the word "Waterloo." Colonel John Clark-Kennedy, C.B., died in Egypt in 1867. The remains of Mrs Clark-Kennedy were interred in the family burying ground in St Michael's Churchyard, Dumfries.'
