Born Charlotte Frances Frederica Seymour on 28 September 1835, she was the daughter of Frederick Charles William Seymour and his wife, Lady Augusta née Hervey. On 8 July 1858 she married John Poyntz Spencer (1835-1910), 5th Earl Spencer, but the union did not produce any heirs. She died on 31 October 1903, at the age of 68, at St James’s Place, London. She was buried on 5 November 1903 at Althorp in Northamptonshire.
